(A) Differentiated 3T3L1 cells and (B) HepG2 cells were treated without (N) or with 1 μM of BI-78D3 (BI), Valproic acid (VPA), Acebutolol (AC), lithium chloride (LICL), Niacinamide (NAA) or Vincristine sulfate (VCR) for 1 h respectively followed by stimulation with TNFa 20ng/ml for 15 min. The levels of phosphorylated JNK, total JNK and actin were detected by western blot. Band intensities were measured by ImageJ program and then normalized based on TNFα-only treated cells. The Bar graph shows mean folds of phospho-JNK versus JNK. The experiment was repeat three times independently. Statistical analysis was assessed by one-tailed unpaired t-test; n=3; *p<0.05, **p<0.01, ***p<0.001 vs. TNFα-only treated cells.
