(A) IgE-sensitized BMMCs were preincubated for 1 h with the indicated concentrations of britanin or Bay 61-3606 and stimulated with DNP-HSA for 15 min. Cytosol fractions were prepared for the determinations of IKKα/β and IκBα phosphorylation and total IκBα levels by immunoblotting. (B) IgE-sensitized BMMCs were pretreated with britanin or Bay 61-3606 for 1 h and stimulated by DNP-HSA for 30 min. Nuclear extracts were then analyzed by immunoblotting for the NF-κB-p65 subunit. The relative nuclear p65/lamin B protein ratios were determined by measuring immunoblot band intensities using a scanning densitometer. The results from three separate experiments as relative ratios (%) are represented. Values are shown as the mean ± S.D. from three independent experiments, **p<0.01 and ***p<0.001 versus the IgE/Ag-induced group.
